Fluorescence correlation spectroscopy FCSmonitors the relative fluorescence fluctuations in a small confocalvolume element light green in figure which is typically less than1 femtoliter The fluorescence photons emitted from molecules inthis volume element pass through a pinhole and are detected by ahighly sensitive detector The signaltonoise ratio achieved bythis method is very high since signal interference from scatteredlaser light background fluorescence and Raman emission can belargely eliminated This allows measurements at the single moleculelevel    See Answer
